RNA isolation, total RNA kit, E-Z 96®

Descrição: The E-Z 96® Total RNA Kit is designed for isolation of total cellular RNA from up to 5×10⁶ cultured cells or soft tissues. This kit can process single or multiple samples in less than 60 minutes. Utilising HiBind® silica plate technology, the need for phenol/chloroform extractions, CsCl gradient ultracentrifugation, and precipitation with isopropanol or LiCl are eliminated. Samples are lysed in a denaturing lysis buffer which inactivates RNases. Binding conditions are adjusted and the lysate is transferred to a 96-well HiBind® RNA Plate where the RNA is purified via three wash steps. High quality RNA is eluted in RNase-free water. RNA purified using the E-Z 96® Total RNA method is ready for applications such as RT-PCR, qPCR, differential display, microarrays, and other downstream applications.

Descrição: Cytokines for polarizing T cells into T regulatory cells. T regulatory cells (Tregs) are a subset of T cells that suppress immune function. They are characterized by the expression of CD4, CD25 and Foxp3 and are known to produce immunosuppressive cytokines TGF-β 1 and IL-10. Tregs can be cultured from naive CD4+ T cells by culturing them in the presence of IL-2 and TGF-β 1 with CD3 and CD28 stimulation. In some cases cells are also cultured with neutralizing antibodies to IL-4, IFNγ and IL-12 and/or retinoic acid.

Descrição: In 1976, Guilbert and Iscove demonstrated that precursor cells of erythrocytes and macrophages could be cultured in a reduced-serum medium supplemented with albumin, transferrin, lecithin, and selenium. Iscove's medium is a modification of Dulbecco's Modified Eagle's Medium (DME) containing selenium, additional amino acids and vitamins, sodium pyruvate, HEPES buffer, and potassium nitrate instead of ferric nitrate. Further studies have demonstrated that Iscove's medium will support murine B lymphocytes, haemopoietic tissue from bone marrow, B-cells stimulated with lipopolysaccharide, T-lymphocytes, and a variety of hybrid cells.
