Descrição: Um excelente meio selectivo para a detecção da enterobactéria <i>E. sakazakii</i> no leite em pó em geral e nos leites em pó para lactentes. O meio E.sakazakii ChromoCult ® permite o bom crescimento e a forte coloração das colónias de <i>E.sakazakii</i>. A adição de inibidores e a temperatura de incubação de 44 °C reduzem largamente o crescimento da maior parte da flora acompanhante de bactérias gram-positivas e gram-negativas. A E. sakazakii é beta-D-glucosidase positiva e cresce neste meio na forma de colónias verde-azuladas. Incubação: 24 ±2 horas a 44 ±1 °C. A temperatura de incubação tem uma forte influência na sensibilidade e na selectividade deste método. Temperaturas superiores a 45 °C inibem o crescimento da <i>E. sakazakii.</i> Temperaturas inferiores a 43 °C reduzem a inibição das bactérias acompanhantes.

Descrição: Chromogenic media offer a method for rapid identification of characteristic bacterial enzymes. Merck offers a wide range of solid and liquid culture media that also permits the fluorescence detection of <i>E. coli</i> under the trade name Fluorocult®. Fluorocult® culture media have the same components as the standard media and additionally the substrate MUG. Some of the media also contain tryptophan as the substrate for a possible indole reaction to further confirm <i>E. coli</i> presence. The Fluorocult® method can produce a positive result within just 24 to 48 hours. In the case of the chromogenic media, a direct identification, using the characteristic colony colouring on the culture medium itself is possible without the application of further additives. A differentiation of positive single colonies is possible, even when high microbial counts are present.
