Descrição: This T4 DNA ligase catalyzes the formation of a phosphodiester bond between juxtaposed 5'-phosphate and 3'-hydroxyl termini in duplex DNA or RNA. The enzyme repairs single-strand nicks in duplex DNA, RNA, or DNA/RNA hybrids. It also joins DNA fragments with either cohesive or blunt termini, but has no activity on single-stranded nucleic acids.

Descrição: The rGRFT is an Escherichia coli recombinant of carbohydrate-binding protein (Lectin) derived from Griffithsia sp. This lectin preferably binds to a [Manα(1-2)Man] glycan epitope.
The LecBeads is a lectin-immobilized agarose which is capable of capturing glycoproteins via binding to its specific epitope(s), and it is prepared by a covalent coupling method between lectin and agarose.
The Lectin-catch method using LecBeads (as shown in Application tab) is available for selective separation of glycoconjugates (glycoproteins, glycolipids, and so on). This rGRFT-LecBeads (1 ml gel) shows a binding capacity of about 6 mg or more bovine Ribonuclease B [RNase B] through its binding to a terminal [Manα(1-2)Man] glycan epitope on RNase B.

Descrição: Streptavidin Resin is designed for the single step small and large scale affinity purification of proteins and antibodies with a biotin tag. Biotin, a 244 Da vitamin (Vitamin H) molecule, exhibits an extraordinary binding affinity for avidin (Ka=10¹⁵ M⁻¹) and streptavidin (Ka=10¹⁵ M⁻¹). Biotin and (strept)avidin interaction is rapid and once the bond is established it can survive up to 3M guanidinehydrochloride and extremes of pH. Biotin-avidin bonds can only be reversed by denaturing the avidin protein molecule with 8M guanidine-hydrochloride at pH 1,5 or by autoclaving. The biotinylated molecules are efficiently probed with avidin or streptavidin conjugated to reporter molecules, such as peroxidases or phosphatases.

Concanavalin A agarose

Descrição: Concanavalin A (Con A) agarose consists of Con A coupled to 6% agarose by the cyanogen bromide method. Con A is a tetrameric metalloprotein lectin isolated from <i>Canavalia ensiformis</i> (jack bean). It is used for the purification of glycoproteins, polysaccharides and glycolipids as it binds molecules containing α-D-mannopyranosyl, α-D-glucopyranosyl and sterically related residues. It has also be used in other application areas including purification of enzyme-antibody conjugates, purification of IgM and separation of membrane vesicles. As stated above, it is a metalloprotein and to maintain its binding characteristics the presence of both Mn²⁺ and Ca²⁺ is essential. Each subunit of Con A utilises one calcium and one manganese ion and these cations can be removed under acidic conditions abolishing the carbohydrate-binding activity.

Descrição: The use of peptides for the generation of antibodies against specific peptides has become an essential tool in proteomic research. A crucial step in using peptides as antigens is their coupling to carrier proteins. Peptides are designed to be good epitopes by themselves; however they fail to elicit a strong antibody response. The reason for this is that the epitopes are able to bind to the B-cells, but are too small to contain additional motifs required to associate with class II molecules and T-cell receptors. The carrier proteins, once coupled to the peptides, provide these additional motifs and generate a strong antibody response. All three carrier proteins are also offered as activated carrier proteins, named as ActiveHOOK™.
