Descrição: The product encoded by this gene is involved in the maintenance of iron homeostasis, and it is necessary for the regulation of iron storage in macrophages, and for intestinal iron absorption. The preproprotein is post-translationally cleaved into mature peptides of 20, 22 and 25 amino acids, and these active peptides are rich in cysteines, which form intramolecular bonds that stabilize their beta-sheet structures. These peptides exhibit antimicrobial activity. Mutations in this gene cause hemochromatosis type 2B, also known as juvenile hemochromatosis, a disease caused by severe iron overload that results in cardiomyopathy, cirrhosis, and endocrine failure. [provided by RefSeq, Jul 2008].

Descrição: Bone morphogenic proteins (BMPs) are members of the TGF Beta superfamily. BMPs are involved in the induction of cartilage and bone formation. In vivo studies have shown that BMP-2 (also designated BMP-2A) and BMP-3 can independently induce cartilage formation. Smad3 association with the TGF Beta receptor complex and Smad1 translocation to the nucleus are observed after the addition of BMP-4 (also designated BMP-2B), suggesting that BMP-4 may play a role in activation of the Smad pathway. BMP-5, BMP-6 and BMP-7 all share high sequence homology with BMP-2, indicating that they each may be able to induce cartilage formation. BMP-8 (also designated OP-2) is thought to be involved in early development, as detectable expression has not been found in adult organs.

Descrição: SPPL2b is a 592 amino acid multi-pass membrane protein that contains one protease associated domain and belongs to the GXGD family of aspartic proteases. Localized to the membrane of the Golgi apparatus and to lysosomes, endosomes and the plasma membrane, SPPL2b functions as an intramembrane protease that specifically cleaves the transmembrane domain of TNF? Once cleaved, TNF?releases its intracellular domain, thus triggering the immunity-related expression of cytokines (also known as interleukins) which are used in various pathways throughout the body. SPPL2b also interacts with and catalyzes the intramembrane proteolysis of ITM2B (integral membrane protein 2B), a protein that, when mutated, is associated with dementia. Four isoforms of SPPL2b exist due to alternative splicing events.

Descrição: PDZ-GEF1 is a 1,499 amino acid cell membrane protein that functions as a guanine nucleotide exchange factor for Rap 1A, Rap 1B and Rap 2B GTPases. Expressed at highest levels in brain, PDZ-GEF1 is found at low levels in placenta, heart, lung and kidney, and undergoes post-translational phosphorylation following DNA damage. PDZ-GEF1 interacts with MAGI-2 and contains one Ras-GEF domain, a Ras-associating domain, one PDZ (DHR) domain, a single N-terminal Ras-GEF domain and a cyclic nucleotide-binding domain. The gene encoding PDZ-GEF1 maps to human chromosome 4q32.1.
