T4 DNA Ligase catalyses the joining of two cohesive- or blunt-ended strands of DNA between the 5'-phosphate and the 3'-hydroxyl groups of adjacent nucleotides. T4 DNA Ligase is provided with 10X reaction buffer: 300 mM Tris-HCl (pH 7,8 at 25 °C), 100 mM MgCl₂, 100 mM DTT and 10 mM ATP.
- Use with 5', 3' or blunt-ended dsDNA
- Qualified for blue/white cloning
- Providing a higher level of quality control for enzymes used in cloning applications
- It can catalyse the joining of RNA to either a DNA or RNA strand in a duplex molecule
- Will not join single-stranded nucleic acids
Storage buffer: 10 mM Tris-HCl (pH 7,4 at 25 °C), 50 mM KCl, 1 mM DTT, 0,1 mM EDTA and 50% glycerol.
Unit definition: 0,01 Weiss unit of T4 DNA Ligase is the amount of enzyme required to catalyse the ligation of greater than 95% of 1 μg of λ/HindIII fragments at 16 °C in 20 minutes.
