Descrição: Recognizes a 14 kDa protein, identified as S100A9 (also known as Calgranulin B or MRP-14); expressed by granulocytes, monocytes and by tissue macrophages.The protein encoded by this gene is a member of the S100 family of proteins containing 2 EF-hand calcium-binding motifs. S100 proteins are localized in the cytoplasm and/or nucleus of a wide range of cells, and involved in the regulation of a number of cellular processes such as cell cycle progression and differentiation. Altered expression of this protein is associated with the disease cystic fibrosis. This MAb reacts with neutrophils, monocytes and macrophages, and has been shown as an important marker for identifying macrophages in tissue sections. Descrição: Recognizes a phosphorylated protein of 228 kDa, identified as nuclear mitotic apparatus protein (NuMA). Its epitope is resistant to phosphatases. NuMA is intra-nuclear protein and present in nucleus during interphase. At the onset of mitosis, it redistributes from the nucleus to two centrosomal structures that later will become part of the mitotic spindle pole. After anaphase, the protein redistributes from the spindle polar region into reforming nucleus. NuMA is an essential protein during mitosis for the terminal phases of chromosome separation and/or nuclear reassembly. Recently a study shows that NuMA is cleaved to a 180 to 200 kDa during apoptosis. Chromosomal translocation of this gene with the RARA (retinoic acid receptor, alpha) gene on chromosome 17 has been detected in patients with acute promyelocytic leukemia.

In the epidermis, expression of CK10 strictly parallels the extent of differentiation; it is absent in the basal layer, appears in the first suprabasal layers and increases in concentration towards the granular layer. However, CK10 is rarely detected in early stages of vulvar squamous carcinomas (tumors less than 2 cm, clinical stage I) regardless of the tumor grade. In larger and more advanced tumors (greater than 2 cm, clinical stages II and III), CK10 is detected very frequently. Expression of CK10 is related to maturation of malignant keratinocytes, being preferentially detected in more-differentiated parts.

Markers of myeloid cells are useful in the identification of different levels of cellular differentiation. It reacts with early precursor and mature forms of myeloid cells. It is useful for the detection of myeloid leukemias and granulocytic sarcomas. It can be used as a marker of granulocytes in normal tissues or inflammatory processes.G-CSF is a pleiotropic cytokine that influences differentiation, proliferation and activation of the neutrophilic granulocyte lineage.
