Descrição: This lab activity is designed to study highly specific lock-key matching properties of antigen-antibody and how this highly specific interaction can be exploited as a tool for research and analysis. This study involves the use of an immunodiffusion technique in which antigen and antibody are allowed to diffuse in a solid agarose medium. When antigen and antibody meet, antigen-antibody complex is formed, which leads to precipitation. Antigen-antibody precipitate is formed in the zone where the concentration of the two matching pairs reaches an optimal known as the zone of equivalence, which results in formation of a visible opaque precipitate region in agarose medium. Those regions of precipitation can be used for determination of concentration or titer of both antigen and antibody. This Antigen-Antibody Interaction kit is a hands-on study of both Ouchterlony Double Diffusion and Radial Immunodiffusion techniques. This kit also provides additional guidance materials for teaching other types of antigen-antibody interactions concepts such as immunoelectrophoresis and immunoprecipitation.

Descrição: Matrix metallopeptidase 9 (MMP-9) is an enzyme encoded by the MMP9 gene. This protein, which is produced by normal alveolar macrophages and granulocytes, can be activated by 4-aminophenylmercuric acetate and phorbol ester and up-regulated by ARHGEF4, SPATA13 and APC via the JNK signalling pathway in colorectal tumour cells. MMP-9 is involved in the breakdown of extracellular matrix in normal physiological processes, such as embryonic development, reproduction, angiogenesis, bone development, wound healling, cell migration, learning and memory, as well as in pathological processes, such as arthritis, intracerebral hemorrhage, and metastasis.

Descrição: Safe drinking water is vitally important to health. Both pathogenic and harmless bacteria can be found in the guts of mammals and birds. Testing water for every possible type of pathogenic bacteria is slow and costly. Thus, water is tested for a characteristic type of gut bacteria - the coliforms - including the familiar <i>E. coli</i>. Presence of coliforms is an indicator for faecal contamination. In this experiment, students test for coliforms in simulated contaminated water using colour and fluorescent reagents. They can use these same reagents to test water samples from the environment. As an extension activity, a Gram Stain test can be performed on the collected samples.
