Descrição: Protein-S-S-Reductant™ uses TCEP (Tris [2-carboxyethyl] phosphine), a popular alternative to β-mercaptoethanol and DTT (dithiothreitol). TCEP improves stability, increases effectiveness, and reduces proteins over a wider range of pH, including lower acidic pHs. Protein-S-S-Reductant™ completely reduces stable disulfide bonds in less than 5 minutes at room temperature and is compatible with the protein alkylation reactions. This ready to use solution is at a neutral pH and stabilised for long-term storage. Simply supplement Protein-S-S-Reductant™ in place of DTT or β-mercaptoethanol and boil the sample.

Descrição: FLUOR DE LYS® Developer is an essential reagent for signal generation in HDAC or sirtuin assays done with the FLUOR DE LYS® substrate. An appropriate FLUOR DE LYS® family substrate, comprising an acetylated lysine side chain, is first incubated with a sample containing HDAC activity (nuclear or cellular extract, purified enzyme, bead-bound immunocomplex, etc.). Deacetylation of the substrate sensitizes it so that, in a second step, treatment with the FLUOR DE LYS® Developer produces a fluorophore. Typically, an ‘inhibitor stop’ will be included in the 1x Developer dilution (e.g. Trichostatin A) for class I and II HDACs, nicotinamide for sirtuins).

Descrição: Streptavidin Resin is designed for the single step small and large scale affinity purification of proteins and antibodies with a biotin tag. Biotin, a 244 Da vitamin (Vitamin H) molecule, exhibits an extraordinary binding affinity for avidin (Ka=10¹⁵ M⁻¹) and streptavidin (Ka=10¹⁵ M⁻¹). Biotin and (strept)avidin interaction is rapid and once the bond is established it can survive up to 3M guanidinehydrochloride and extremes of pH. Biotin-avidin bonds can only be reversed by denaturing the avidin protein molecule with 8M guanidine-hydrochloride at pH 1,5 or by autoclaving. The biotinylated molecules are efficiently probed with avidin or streptavidin conjugated to reporter molecules, such as peroxidases or phosphatases.

Concanavalin A agarose

Descrição: Concanavalin A (Con A) agarose consists of Con A coupled to 6% agarose by the cyanogen bromide method. Con A is a tetrameric metalloprotein lectin isolated from <i>Canavalia ensiformis</i> (jack bean). It is used for the purification of glycoproteins, polysaccharides and glycolipids as it binds molecules containing α-D-mannopyranosyl, α-D-glucopyranosyl and sterically related residues. It has also be used in other application areas including purification of enzyme-antibody conjugates, purification of IgM and separation of membrane vesicles. As stated above, it is a metalloprotein and to maintain its binding characteristics the presence of both Mn²⁺ and Ca²⁺ is essential. Each subunit of Con A utilises one calcium and one manganese ion and these cations can be removed under acidic conditions abolishing the carbohydrate-binding activity.

Descrição: The use of peptides for the generation of antibodies against specific peptides has become an essential tool in proteomic research. A crucial step in using peptides as antigens is their coupling to carrier proteins. Peptides are designed to be good epitopes by themselves; however they fail to elicit a strong antibody response. The reason for this is that the epitopes are able to bind to the B-cells, but are too small to contain additional motifs required to associate with class II molecules and T-cell receptors. The carrier proteins, once coupled to the peptides, provide these additional motifs and generate a strong antibody response. All three carrier proteins are also offered as activated carrier proteins, named as ActiveHOOK™.
