These receptors have been shown to be involved in long-term potentiation, an activity-dependent increase in the efficiency of synaptic transmission thought to underlie certain kinds of memory and learning. Genetic analysis has identified two proteins that are mutated in some cases of lissencephaly, designated lissencephaly-1 protein (LIS1) and doublecortin. LIS1 displays sequence homology to ?subunits of heterotrimeric G proteins, and doublecortin contains a consensus Abl phosphorylation site. In addition, the DCAMKL1 (doublecortin-like and CAM kinase-like 1) protein shows homology to doublecortin. All three proteins are highly expressed in developing brain and may function together to regulate microtubules involved in neuronal migration. The DCAMKL1 protein encodes a functional kinase that is capable of phosphorylating myelin basic protein and itself, but its kinase activity does not appear to affect its microtubule polymerization activity.

Descrição: Adenylate kinase is an enzyme involved in regulating the adenine nucleotide composition within a cell by catalyzing the reversible transfer of phosphate group among adinine nucleotides. Three isozymes of adenylate kinase have been identified in vertebrates, adenylate isozyme 1 (AK1), 2 (AK2) and 3 (AK3). AK1 is found in the cytosol of skeletal muscle, brain and erythrocytes, whereas AK2 and AK3 are found in the mitochondria of other tissues including liver and heart. AK1 was identified because of its association with a rare genetic disorder causing nonspherocytic hemolytic anemia where a mutation in the AK1 gene was found to reduce the catalytic activity of the enzyme. Expression of TUSC2 protein is absent or reduced in the majority of lung cancers and premalignant lung lesions. Restoration of TUSC2 function in 3p21.3-deficient non-small cell lung carcinoma cells significantly inhibits tumor cell growth by induction of apoptosis and alteration of cell cycle kinetics. TUSC2 may induce apoptosis through the activation of the intrinsic mitochondrial-dependent and Apaf-1-associated pathways and inhibit the function of protein tyrosine kinases including EGFR, PDGFR, AKT, c-Abl, and c-Kit.

Descrição: Calcyon is a single transmembrane protein that interacts with D1 dopamine receptors. Dopamine is a neurotransmitter that regulates synaptic transmission involved in learning and memory. D1 receptors, the most abundant dopamine receptor in the central nervous system, appear to modulate the activity of D2 dopamine receptors, mediate various behavioural responses, and regulate neuron growth and differentiation. Calcyon is present in neuronal cell bodies and processes of the cortex and hippocampus, and it is especially abundant in pyramidal neurons. Interaction of Calcyon with D1 receptors results in a release of intracellular calcium.
